Vibrio vaccination and protective effect of serum transfer in cod
Begrunnelse for etterevalueringen
For enkelte torsk forventes forsøket å være betydelig belastende.
Etterevaluering
This FOTS project is a follow-up and extension of previous experiments where the main aim is to investigate how anti-vibrio IgM works in protective immunity in cod. Due to technical issues and fish health issues in general in the facility, we did not manage to achieve some of the main aims, such as whether fish with naturally high anti-vibrio IgM levels have better survival; whether passive transfer of immune serum also protects infectious challenge via the i.p. route; and testing the efficacy of a novel component vaccine.
We did find that the half life of IgM in cod is about 4 days, with some biological variation.
In total 238 fish were used in the experiments covered by this FOTS. Of these, the severity was judged to be n=9 for mild, n=196 for moderate, and n=33 for severe. We used 16 more fish than originally planned to test the correlation between specific antibody level and immune protection. The number of fish used was otherwise relatively similar to the original plan.
There is virtually no cod cell line or other specific reagents available for the study of cod. We conducted vaccination efficacy experiments that are very difficult to simulate by in vitro models, especially given the unique immune system of cod.
