Blood sampling and termination of Diversity outbred mice

1. This project is part of a EU-funded study (Horizon 2020 – Project#. 848099) where the overall goal is to identify novel biomarkers and genetic variants related to pain sensitivity, inflammation and cardiovascular risk. Initially all animal housing was planned abroad. When finished, personnel and expertise would travel to successfully harvest tissues and organs is such a way that is suitable for downstream analysis. Due to the ongoing pandemic, transportation of the mice to us remains the only option. The mice will be housed for a maximum of 4 – 6 weeks with atherogenic diet and water ad libitum before termination.
2. One blood sample (50 µL) through a pinprick to the tail or calf vein will be taken before termination. A mild stress can be expected, but the initial pain will be short-lived.
3. This project will be a part of a larger EU-funded project with the overall aim of identifying novel biomarkers and gene variants related to pain sensitivity, inflammation and cardiovascular risk.
4. In total, 300 male and female Diversity Outbred mice will be used in this study for maximum genetic diversity in the study population.
5. The search for biomarkers and genetic variations is impossible to study outside a living organism. This kind of epidemiological studies require large study population for statistic validity. The number of mice included in this study is a compromise between animal welfare, practical work load and statistical validity.
