Study on Piscirickettsia salmonis infection in Medaka fish (Oryzias latipus)

This trial will allow us to establish Medaka fish as a new model system to study Piscirickettsia salmonis pathogenesis.

This trial will develop an infection model for the disease Salmonid Rickettsial Syndrome (SRS) caused by P. salmonis for the first time in Medaka. This fish model has been well studied to bring new insights into disease development and innate immune responses of marine fish.
In order to determine the effect of P. salmonis in Medaka fish, we will use both genogroups identified for the bacteria, LF89 and EM90 in single and co-infection experiments. We plan to compare virulence and disease dynamics during infection for the two genotypes. Also, by expression gene analyses we can elucidate if there is any change in the pathogen and in the host expression profile when the bacteria infect alone or together, which may suggest a synergistic effect under in vivo conditions in this model system.

We will use 300 Medaka fish (Oryzias latipus) for the trial, and an additional 20 Medaka fish for the pilot

Begrunnelse for etterevalueringen

The medaka model fish cannot be used in replacing salmon for studying piscirickettsiosis.
We used 37 out of the 320 animals applied for the experiment because we did not continue with the main trial after ending the pilot experiment.
We could use the planned medaka model and therefore we could not improve any of these parameters.
