Salmonid rickettsial septicemia (SRS) is a devastating disease caused by the intracellular pathogen Piscirickettsia salmonis. SRS is a major concern of today’s aquaculture industry causing major economic losses due to the high mortality rates in Atlantic salmon. Development of a vaccine against this pathogen is challenging because of the intracellular nature of the pathogen.
The purpose of this study is to investigate the use of P. salmonis MVs and LPS as vaccines/adjuvants against SRS in adult zebrafish. For the LPS trial, the study will be divided into two parts with a required number of 200 adult zebrafish. The first part of the study will evaluate the effect of the increasing LPS dose while the second part will evaluate the toxicity of the LPS isolated from different P. salmonis strains.
The use of MVs has been tested before in adult zebrafish, zebrafish embryos and cell cultures. The number of fish used in this experiment is therefore based on the previous experiments to minimize unnecessary use of experimental animals. To reduce the pain of fish used in the study, anesthesia will be used during the duration of the experiments. Fish with any clinical signs of SRS, will be removed from the trail and anesthetized.
