Identification of immunocharacteristics after UVT/Botox in neoplastic/gastric tissue in INS-GAS mice
Previous studies in Duan Chens group have investigated the effect on the INS-GAS tumor metabolome and inflammation-related signaling pathways after unilateral truncal vagotomy (UVT) or local injection of Botulinum toxin A (Botox), (Ref. Zhao et al., Sci Trans Med, 2014). The purpose of this study is to investigate the long term effect of UVT or Botox on gastric/tumor immune system in INS-GAS mice with the aim to identify the "immune fingerprint". This study could provide insight of the immune responses involved in neoplastic tissue with or without UVT/Botox treatment.
After establishment of the immuno-fingerpint, we would like to carry out combination treatments with denervation, chemotherapy, metabolite inhibitors and immunotherapy. Therefore we would like to apply: at first for more breeding of INS-GAS mice (476 mice); secondary perform the combination therapy on these INS-GAS mice.
Our previous experience with similar experiments has been published in our paper "Denervation Suppresses Gastric Tumorigenesis” in Science Translational Medicine 2014 and has been recognized as a breakthrough and since been cited 52 times including Cell (3 times), Nature Reviews Cancer (2 times), Nature Reviews Gastroenterology, Cancer Cell, PNAS, etc. This research has been followed by NRK Dagsrevyen, BBC, The Time, The Telegraph, Yahoo, Fox, Daily Mail, US Daily, Medscape, etc. Altmetric has tracked 5,336,996 research outputs across all sources so far. The article is in the 99th percentile: it's in the top 5% of all research outputs ever tracked by Altmetric (https://www.altmetric.com/details/2612911#score). Additionally we would like to point out that by ensuring good internal control groups and sufficient group numbers we aim to act in accordance with the 3R principle and to produce high quality data that will lead to less need for repeated experiments and errors in the future.
