Immune-instructive biomaterials for modulating bone regeneration in aged and osteoporotic rats

1 Purpose
The repair and treatment of large bone defects following fractures, ablative surgery or pathological bone changes presents often a clinical challenge in patients with compromised healing, such as aged or osteoporotic patients. Therefore, adjunctive methods to control inflammation to promote bone healing in these patients are needed. The aim is to test the potential of scaffolds functionalized with or without immunomodulatory molecules derived from plants delivering mesenchymal stem cells on bone regeneration. This will be tested in rat calvarial bone defects using Lewis rats after the induction of osteoporosis. The autogenous bone graft will be used as a “gold standard” bone regeneration therapy.

2 Distress
Rats will undergo the ovariectomy and calvaria bone defects under general anesthesia and analgesia will be administered pre and post operatively.

3 Expected benefit
In vitro experiments have been extensively studied with regard to the Gal RG-I alone and in addition to being incorporated in scaffolds. The need to evaluate its efficacy in regenerating bone in a immunocompromised or inflammatory environment is essential for clinical applications. Our results will help confirm the synergistic effect of mesenchymal stem cells and our immunomodulatory molecules to promote bone regeneration. this will have a great impact in enhancing bone regenerative therapies in an aging and/or osteoporotic population.

2 defects will be created in each rat, to reduce the total number of rats used. All surgeries will be undergone under general anesthesia and analgesia will be administered.
