Characterization of Ythdf1 and Ythdf3 in mice with reporter genes
I) The purpose of the experiment/project:
To study the role of Ythdf1 and Ythdf3 in the mouse development.
II) The expected adverse effects on the animals:
These animals are not supposed to have any difficulties compared to wildtype mice. They have a small knock in sequence at the end of Ythdf1 and Ythdf3 respectively. Our experiments will be mainly focused on early stages of development, so the animals will not suffer any pain or discomfort during their lives.
III) The expected scientific benefits or benefits for society:
The more we research on methyl-6-adenosine mRNA modifications the more we can relate it to important processes like development or cancer. Ythdf1 and Ythdf3 have shown already a role in these activities and we want to throw some light in their role in development and neurogenesis.
IV) The number of animals and species:
We would like to use around 512 mice (Mus musculus) of the strain C57BL/6N.
V) How will the requirements for 3R be accomplished by the experiment/project:
Our experiments require the use of animals, but we will help reduce the number of them that we need to use by developing different cell lines from them to continue with the experiments. We have been working with this strain in the past and this experience will help us refine not only the experiments but also any distress the animals could experience.
